Note1 : The cells were transfected with 50nM of the target siRNA, 48h prior to live cell imaging (n= 336 cells, from 2 independent experiments).
Note2 : 51% of cells showed chromosome congression defects
Note3 : 25% of the cells died in mitosis without completing chromosome alignment; 3% of the cells died in the subsequent interphase
Note4 : The duration of mitosis was 98±217 min (mean±sd, determined by measuring the time between nuclear envelope breakdown (NEBD) to anaphase onset (AO)).

Protein lysates obtained after siRNA were immunoblotted with an antibody specific to each protein of interest. Alfa-tubulin was used as loading control.
