Note1 : The cells were transfected with 50nM of the target siRNA, 24h prior to live cell imaging (n= 79 cells, from 1 experiment).
Note2 : 51% of cells showed chromosome congression defects
Note3 : 8% of the cells died in mitosis without completing chromosome alignment; 37% of the cells died in the subsequent interphase

Protein lysates obtained after siRNA were immunoblotted with an antibody specific to each protein of interest. Vinculin was used as loading control.
