Note1 : The cells were transfected with 50nM of the target siRNA, 24h prior to live cell imaging (n= 325cells, from 2 independent experiments).
Note2 : 42% of cells showed chromosome congression defects
Note3 : 13,8% of the cells died in mitosis without completing chromosome alignment; 0% of the cells died in the subsequent interphase
Note4 : The duration of mitosis was 89±147 min (mean±sd, determined by measuring the time between nuclear envelope breakdown (NEBD) to anaphase onset (AO)).

Protein lysates obtained after siRNA were immunoblotted with an antibody specific to each protein of interest. GAPDH was used as loading control.
